Clean Room Construction: 2026 Industry Requirements

Clean room construction must begin with the contamination risk—not a copied ISO class. Pharmaceutical suites, semiconductor fabs, battery dry rooms, and hospital isolation rooms use controlled air, but need different pressure, humidity, finishes, utilities, and tests. The comparison below helps engineering and procurement teams write a usable request for quotation.

Pharmaceutical and Biotech Clean Room Construction

Separate Sterile, Non-Sterile, Biological, and High-Potency Process Requirements

Sterile filling needs protected critical zones, qualified airflow, environmental monitoring, and a contamination-control strategy. EU GMP Annex 1 has applied since August 25, 2023, with its deferred provision applying since August 25, 2024; FDA aseptic guidance supports 21 CFR Parts 210 and 211.

Non-sterile tablets may need dust containment instead of aseptic protection. Biological production adds segregation, while potent compounds may require negative-pressure isolators and dedicated exhaust. Protection improves, but energy, cleaning, and change-control costs rise.

Define Cleanroom Grades, Pressure Cascades, Hygiene Zones, and Validation Scope

ISO 14644-1:2015 classifies airborne particle cleanliness, but pharmaceutical acceptance also depends on viable monitoring and GMP controls. A 10–15 Pa room-to-room pressure step is a common starting point, adjusted for doors and containment.

In our experience, late changes to personnel flow and equipment footprints create more rework than panel selection. Freeze the URS, material routes, and FAT-SAT-IQ-OQ-PQ scope before cleanroom construction contracts are released.

Semiconductor, Electronics, and Battery Facility Requirements

Control Particles, Airborne Molecular Contamination, ESD, Temperature, and Humidity

Advanced fabs may combine ISO 3–5 zones, mini-environments, ESD flooring, chemical filtration, and tight temperature control. SEMI F21 classifies airborne molecular contamination, while 2024 IFC Chapter 27 covers semiconductor hazards involving flammable, pyrophoric, toxic, and corrosive materials.

Commercial cleanrooms generally follow locally adopted IBC and IFC provisions, not the residential IRC. This cleanroom construction protects yield but consumes substantial utilities; compare fan power, AMC media life, vibration, and filter coverage—not only ISO class.

Integrate Equipment Cleanliness, Material Compatibility, Utilities, and Dry-Room Zones

Battery cleanroom manufacturing may need less particle control than wafer fabrication but much lower moisture. DOE and Argonne references describe dry rooms below 100 ppm moisture and supply-air dew points around -40°C to -63°C.

From the projects we’ve completed, the costliest error is treating a dry room as comfort HVAC. People, doors, compressed air, and incoming materials add moisture; without a load model, the desiccant system may miss specification.

Select ISO Classes and Pressure Direction from Product and Patient Risks

Not every process needs ISO classification. FDA food rules under 21 CFR Part 117 focus on sanitation and preventive controls. FDA’s medical-device QMSR became effective February 2, 2026, and incorporates ISO 13485:2016.

For cleanroom manufacturing of implants or sterile disposables, ISO 7–8 backgrounds are common starting points. Food, cosmetics, and general laboratories may only need hygienic zoning; ISO 22716 covers cosmetic GMP for production, control, storage, and shipment.

Specify Hygienic Materials, Cleaning Methods, Monitoring, and Certification Tests

Specify coved floors, sealed penetrations, low-shedding panels, compatible sealants, and validated cleaning methods. ASHRAE/ASHE Standard 170 governs healthcare ventilation relationships; airborne infection isolation rooms remain negative to adjacent spaces.

Ask clean room manufacturers to price particle classification, HEPA integrity, airflow, recovery, pressure mapping, temperature and humidity mapping, and microbial qualification separately. This exposes incomplete low bids.
